Lipophilicity and Biomimetic properties supporting drug discovery

The course explains the answers to the following questions:

What is lipophilicity and why it is important?
What types of lipophilicity to use in drug discovery (octanol/water, chromatographic and biomimetic lipophilicity)?
How can octanol/water partition, chromatographic lipophilicity and biomimetic HPLC properties be measured?
How can various lipophilicity measurements be compared by the solvation equation approach?
How does lipophilicity depend on the pH?
How can the measured lipophilicity (PFI) be used in early drug discovery to establish compound ranking and progression?
Structure- lipophilicity relationships
What is volume of distribution, tissue plasma partition and why is it important?
What is unbound volume of distribution and drug efficiency?
Which biomimetic properties of compounds affect in vivo distribution?
How can we use these properties at an early stage of the drug discovery process? (Case histories will be discussed)
How in vivo distribution profiles affect the estimated clinical dose?


The course is suitable for analytical chemists, medicinal chemists and physicochemist working in a drug discovery environment.

The course is designed for small groups (max 15) and to be interactive allowing questions and answers and demonstrating case histories. Attendees will be given course material and a certificate of attendance.

What is aqueous solubility?
What are the conditions that affect the measured solubility values?
How can solubility be measured in the early drug discovery stage?
How does solubility depend on the pH?
What are the causes of solubility enhancement in simulated intestinal or lung fluids?
How does solubility in various media depend on the chemical structure?
Why solubility is important in drug discovery and how the solubility data can be used for compound design, selection and progression?
What is permeability and why is it important in early drug discovery?
What is artificial membrane permeability and what is the difference compared with cell permeability?
How permeability depends on the pH and various sink conditions?
How can we use permeability and solubility to estimate oral absorption?
